Abstract:
The glucose and fructose content in raw and white sugars is determined using High Performance Anion-Exchange Chromatography.
The method is suitable for contents of glucose and fructose between 0.005 g/100 g and 0.15 g/100 g in the sample.
Sugars are separated using a pellicular anion exchange column and detected using a pulsed amperometric detector (PAD).
Comparison with a lactose standard solution provides identification and quantification.
For raw sugars with glucose or fructose levels between 0.08 g/100 g and 0.23 g/100 g the absolute difference between two results obtained under repeatability conditions should not be greater than 0.02 g/100 g and under reproducibility conditions not greater than 0.03 g/100 g.
For white sugars with glucose or fructose levels between 0.004 g/100 g and 0.017 g/100 g the absolute difference between two results obtained under repeatability conditions should not be greater than 0.002 g/100 g.
For white sugars with glucose levels between 0.004 g/100 g and 0.014 g/100 g the absolute difference between two results obtained under reproducibility conditions should not be greater than 0.006 g/100 g.
For white sugars with fructose levels between 0.004 g/100 g and 0.017 g/100 g the absolute difference between two results obtained under reproducibility conditions should not be greater than 0.008 g/100 g.
